Epitalon Protects Against Post-Ovulatory Aging-Related Damage of Mouse Oocytes In Vitro

Scientific Summary

This in vitro study evaluated Epitalon, also known as Epithalon, in a mouse oocyte aging model. The researchers added Epitalon to the culture medium and assessed oocyte quality across 6, 12, and 24 hours of post-ovulatory aging. Epitalon treatment reduced intracellular reactive oxygen species, decreased spindle defects and abnormal cortical granule distribution, and increased mitochondrial membrane potential and mitochondrial DNA copy number. The authors reported that these effects were associated with reduced apoptosis during the in vitro aging window and suggested that Epitalon may delay oocyte aging through modulation of mitochondrial activity and oxidative stress.

Plain English Interpretation

This was a controlled laboratory study, not a human-use trial. In cultured mouse egg cells, Epithalon was associated with lower oxidative stress and better preservation of several cellular quality markers during aging outside the body. The main research takeaway is that Epithalon may interact with mitochondrial and oxidative-stress pathways in cell-culture conditions.

This study investigated AEDG peptide, identified as Epitalon/Epithalon, in human gingival mesenchymal stem cells. The researchers reported increased synthesis and mRNA expression of neurogenic differentiation markers including Nestin, GAP43, β Tubulin III, and Doublecortin. Molecular modeling suggested that AEDG may bind to histone H1 regions that interact with DNA, providing a proposed epigenetic mechanism for changes in neurogenesis-related gene expression and protein synthesis.

Plain English Interpretation

This cell-based study looked at how Epithalon may affect gene-expression programs tied to neuronal differentiation. This in vitro study examined several short peptides, including AEDG, in aging skin fibroblast cultures. The researchers assessed markers related to proliferation, regeneration, aging, apoptosis, and extracellular matrix remodeling. Across the studied peptides, the authors reported reduced MMP-9 synthesis, increased Ki-67 and CD98hc expression, and, for AED and AEDG, suppression of caspase-dependent apoptosis that increased during cell-culture aging.

Plain English Interpretation

This laboratory study tested how short peptides affected aging-related behavior in cultured skin fibroblast cells. AEDG, the tetrapeptide commonly referred to as Epithalon/Epitalon, was associated with changes in markers of cell proliferation, tissue-remodeling activity, and programmed cell-death signaling. The findings support Epithalon’s use as a research compound for studying cellular aging pathways in vitro.

Verifiable certificates

Beware of fraudulent laboratories

Not only must one remain vigilant about potentially fraudulent certificates, one must also be aware that there are third-party laboratories whose results cannot be considered reliable or scientifically valid. Unfortunately, there is significant evidence to suggest that one of the most popular third-party testing labs does not use scientifically sound methodologies and, in some cases, has fabricated results. There isn’t an easy remedy for this problem, but when labs are particularly bad, there tend to be a lot of discussion threads on various social platforms.

Interpreting Endotoxin Results (via TrustPointe)

Our friends at TrustPointe have provided the following detailed explanation to help interpret the results of the endotoxin testing.

We use the Charles River Endosafe PTS system to test for bacterial endotoxins following USP <85> guidelines:

  • USP <85> Bacterial Endotoxin Result: <x.xx EU/mL
    Because it’s reported as “<x.xx” this indicates the test did not detect endotoxin above the detection limit of the cartridge.
  • If a result is above the limit of detection of the cartridge, it will be reported as a number (without the “<“).

The following are suitability parameters that verify the system was working properly and the sample prep dilution is appropriate for accurate results. Peptides often interfere with endotoxin detection due to their tendency to bind or mask endotoxins, which can lead to inaccurate low results. To overcome this, samples are typically tested at a large dilution to reduce matrix interference and ensure reliable recovery and detection in compliance with USP <85>.  If the dilution is not correct, the run will fail suitability and we’ll need to adjust the dilution to ensure accurate results. We provide the suitability data to customers for transparency and so they can be confident in the results.

USP <85> Sample CV %:

  • CV stands for coefficient of variation, a measure of repeatability.
  • For our lab, CV has to be <25% or the result to be considered valid

USP <85> Spike CV %:

  • This refers to the precision of the positive control (spiked sample).
  • Again, for our lab CV has to be <25% or the result to be considered valid

USP <85> Spike Recovery

  • This tells us how much of the known endotoxin spike was recovered from your sample.
  • The acceptable range is 50–200% per USP <85>
