Study 1: Regenerative and Protective Actions of the GHK-Cu Peptide in the Light of the New Gene Data

Authors: Loren Pickart, Anna Margolina

Source: https://www.mdpi.com/1422-0067/19/7/1987

Scientific Findings

This review summarizes the multifaceted biological effects of GHK-Cu, focusing on its regenerative and protective actions. It details the peptide’s role in stimulating blood vessel and nerve outgrowth, increasing the synthesis of collagen, elastin, and glycosaminoglycans, and supporting dermal fibroblast function. The paper highlights GHK-Cu’s ability to modulate gene expression, affecting a large number of genes related to tissue repair, anti-cancer activity, anti-inflammatory actions, and DNA repair. The authors propose that GHK-Cu’s diverse effects are attributable to its ability to reset the human genome to a healthier state.

Plain English Interpretation

The GHK-Cu peptide is a small, naturally occurring molecule in our bodies that has a wide range of positive health effects. This paper reviews how GHK-Cu helps to repair and protect our tissues, particularly the skin. It works by stimulating the growth of blood vessels and nerves, boosting the production of important structural proteins like collagen and elastin, and supporting the cells that build our skin’s framework. The paper also explains that GHK-Cu can influence thousands of our genes, turning on genes that are good for us and turning off those that are bad. This helps to fight inflammation, protect against cancer, and even repair our DNA, making GHK-Cu a powerful ingredient for skin and hair care products.

Study 2: Synergy of GHK-Cu and hyaluronic acid on collagen IV upregulation via fibroblast and ex-vivo skin tests

Authors: Fangru Jiang, Yanan Wu, Zhe Liu, Minhua Hong, Yi Huang

Source: https://onlinelibrary.wiley.com/doi/10.1111/jocd.15763

Scientific Findings

This study investigated the synergistic effects of GHK-Cu and hyaluronic acid (HA) on collagen synthesis in human dermal fibroblasts and ex-vivo skin models. The researchers found that the combination of GHK-Cu and HA, particularly low molecular weight HA (LMW HA), significantly upregulated the expression of collagen I, IV, and VII. The most potent synergistic effect was observed on collagen IV, with a 25.4-fold increase in cell tests and a 2.03-fold increase in ex-vivo skin tests at a 1:9 ratio of GHK-Cu to LMW HA. The findings suggest that combining GHK-Cu and HA can enhance dermal-epidermal junction (DEJ) health by stimulating collagen IV synthesis.

Plain English Interpretation

This study looked at what happens when you combine two popular skincare ingredients: GHK-Cu and hyaluronic acid (HA). The researchers found that when used together, these two ingredients work as a team to boost the production of collagen, a protein that keeps our skin firm and youthful. They discovered that the combination was especially good at increasing the amount of a specific type of collagen called collagen IV, which is crucial for the connection between the top two layers of our skin. This means that using GHK-Cu and HA together in skincare products could be a great way to improve skin health and fight the signs of aging.

Study 3: GHK-Cu may Prevent Oxidative Stress in Skin by Regulating Copper and Modifying Expression of Numerous Antioxidant Genes

Authors: Loren Pickart, Jessica Michelle Vasquez-Soltero, Anna Margolina

Source: https://www.mdpi.com/2079-9284/2/3/236

Scientific Findings

This paper explores the antioxidant properties of GHK-Cu and its role in preventing oxidative stress in the skin. The study highlights GHK-Cu’s ability to inhibit the formation of reactive carbonyl species (RCS), detoxify toxic products of lipid peroxidation, and protect keratinocytes from UVB radiation. Furthermore, the paper delves into the genetic basis of these effects, revealing that GHK-Cu modulates the expression of numerous antioxidant genes. The authors conclude that GHK-Cu’s antioxidant actions are multifaceted, involving both direct detoxification of harmful molecules and indirect regulation of the skin’s own antioxidant defense systems.

Plain English Interpretation

This paper explains how GHK-Cu acts as an antioxidant to protect our skin from damage. It works in two main ways. First, it directly neutralizes harmful molecules that are created when our skin is exposed to things like sunlight. Second, it helps our skin to protect itself by influencing our genes. The paper shows that GHK-Cu can turn on genes that produce our body’s own natural antioxidants. This two-pronged approach makes GHK-Cu a powerful ingredient for protecting our skin from the damaging effects of the environment and aging.

Storage:

All of our manufacturing partners produce peptides using the Lyophilization (Freeze Drying) process, ensuring products maintain stability for shipping and storage for 12+ months.

In lyophilized form, they are shelf-stable for many weeks. However, for long-term storage, it is recommended to store them in the freezer.

We often hear concerns about the standard “discard after 28 days of first use” disclaimer. Don’t worry, this has nothing to do with studies regarding the efficacy of specific peptides. 28 days is the FDA requirement for producers of multi-use vials to prove their bacteriostatic maintains efficacy. This minimum requirement becomes the de facto standard.

In our experience, if you use proper sterile procedures and refrigerated storage, you can continue sampling from the same reconstituted vial for 3+ months.

Verifiable certificates

When obtaining research peptides, it is essential to validate the authenticity of the Certificate of Analysis (COA). Certificate fraud runs rampant throughout the research peptide supply community. The two most common forms are doctored images and stolen certificates. You can check for these two by making sure the third-party laboratory’s website shows that the certificate belongs to the supplier and the values haven’t been doctored. When obtaining research peptides, it is essential to validate the authenticity of the Certificate of Analysis (COA). Certificate fraud runs rampant throughout the research peptide supply community. TrustPointe Analytics provides a few simple rules for verification:

Beware of fraudulent laboratories

Not only must one remain vigilant about potentially fraudulent certificates, one must also be aware that there are third-party laboratories whose results cannot be considered reliable or scientifically valid. Unfortunately, there is significant evidence to suggest that one of the most popular third-party testing labs does not use scientifically sound methodologies and, in some cases, has fabricated results. There isn’t an easy remedy for this problem, but when labs are particularly bad, there tend to be a lot of discussion threads on various social platforms.

Interpreting Endotoxin Results (via TrustPointe)

Our friends at TrustPointe have provided the following detailed explanation to help interpret the results of the endotoxin testing.

We use the Charles River Endosafe PTS system to test for bacterial endotoxins following USP <85> guidelines:

  • USP <85> Bacterial Endotoxin Result: <x.xx EU/mL
Because it’s reported as “<x.xx” this indicates the test did not detect endotoxin above the detection limit of the cartridge.
  • If a result is above the limit of detection of the cartridge, it will be reported as a number (without the “<“).

The following are suitability parameters that verify the system was working properly and the sample prep dilution is appropriate for accurate results. Peptides often interfere with endotoxin detection due to their tendency to bind or mask endotoxins, which can lead to inaccurate low results. To overcome this, samples are typically tested at a large dilution to reduce matrix interference and ensure reliable recovery and detection in compliance with USP <85>.  If the dilution is not correct, the run will fail suitability and we’ll need to adjust the dilution to ensure accurate results. We provide the suitability data to customers for transparency and so they can be confident in the results.

USP <85> Sample CV %:

  • CV stands for coefficient of variation, a measure of repeatability.
  • For our lab, CV has to be <25% or the result to be considered valid

USP <85> Spike CV %:

  • This refers to the precision of the positive control (spiked sample).
  • Again, for our lab CV has to be <25% or the result to be considered valid

USP <85> Spike Recovery

  • This tells us how much of the known endotoxin spike was recovered from your sample.
  • The acceptable range is 50–200% per USP <85>
